The sides had met in the final of the Barclays Asia Trophy and once again it was Spurs who came out on top to move ahead of Chelsea on goal difference.
Defoe and Palacios both scored inside the opening 14 minutes as Harry Redknapp's side raced out of the blocks at the KC Stadium. It was a first goal in English football for Palacios.
Stephen Hunt pulled one back when his cross from a free-kick bounced in but Defoe broke clear to add a third on the stroke of half-time.
Robbie Keane grabbed the fourth 12 minutes from time and in the dying seconds Defoe completed his hat-trick to bring his tally to seven goals in four matches for club and country.
